On a 66 Mustang. Would a measurement from the center of the groove on the pulleys to the flat engine side of the radiator be an accurate measurement for pulley alignment on all the pulleys?

I've used a steel ruler running alongside the belt between the pulleys to align them. That has always worked best for me.

SInce the engine sits at an angle and the radiator is straight vertical, measuring to the radiator will probably not be very accurate.

Agreed.
Be sure to use the bottom edge of the ruler and not the wide flat part for the alignment.
That little mistake had two edjumicated injuneers jacking with those alignment checks for longer than I'd like to admit :rlaugh:
