No, but there could be circumstances depending on your car...
Every car is different... I've had my ('06) battery disconnected several times and I've never had any issues. However, a buddy of mine ('07) has had his disconnected a few more times than me, but nearly every time it was hooked back up, the car would idle/drive rough for several days. Even after reloading the tune. Come to find out, the computer needs several minutes to relearn the fuel tables and the idle after the battery is disconnected and reconnected. It's stated in the owners manual. What to do is after hooking the battery back up, start the car with the heat/ac off and let it run for a few minutes, then turn on the ac and let it run for a few more minutes. This allows the computer to relearn both with the ac compressor off and on. After doing that, my buddy has never had another idle/drive issue related to battery.
So...no, not required, but might be depending on how your car reacts afterward.
